Abstract
Selective media, PM-1 and PM-2, were developed for isolating Pseudomonas syringae pv. mori (P. s. mori), the causal agent of bacterial blight of mulberry, from soil. The media contain saccharose (PM-1) or sorbitol (PM-2) as a carbon source, and also phenol red, methyl violet, phenethicillin potassium, ampicillin sodium, cetyltrimethylammonium bromide, and cadmium chloride as selective agents.<br>Plating efficiency of P. s. mori was high relatively on the both media, and also all isolates of P. s. magi from various parts of Japan could grow on PM-1 and the majoritty of isolates could grow on PM-2. The colony form of P. s. mori on the media was characteristic and distinguished easily from that of soil-borne microorganisms.<br>With selective media, P. s. mori could be detected when 102 colony forming units were added per gram of non-sterile soil, whereas P. s. mori of 104/g soil could not be detected with the other known media.
