On the activity of fructose-1, 6-diphosphatase in pupal ovaries of the silkowrm, <i>Bombyx mori</i> L.

Description

Fructose-1, 6-diphosphatase (FDPase) which is a key enzyme in the pathway of gluconeogenesis in mammals was surveyed in pupal ovaries of silkworms. FDPase activity was measured according to the method of TAKETA and POGELL (1965). The results are summarized as follows:<br>1) FDPase in pupal ovaries has an optimum activity at pH 8.5. Km value for fructose-1, 6-diphosphate is 5.9×10-5M, but the activity is inhibited by high concentrations of the substrate. Appreciable activation was observed by Mg++ at concentration of 10mM. The enzyme activity is not affected by ATP but inhibited highly by AMP.<br>2) Ovary FDPase remains mainly in the supernatant at 105, 000g in cell fractionation.<br>3) FDPase activities of ovaries increase abruptly from 5-day to 7-day old age and reach maximum at 7-day old age following progressive decrease.<br>4) From these results, it is suggested that FDPase operates in pupal ovaries a few days before adult emergence.
