Biosynthesis of phosphatidic acid in cell-free extract of E. coli was studied by using 32P-orthophosphate and 14C-glycerol. The cells were sonically disrupted and the sonicate was centrifuged at 20, 000 ×g for 25min. When 32P-orthophosphate was incubated with the supernatant, 32P-orthophosphate was incorporated into phosphatidic acid. However, when 14C-glycerol was used, almost all of the radioactivity was recovered as glycerophosphate and the evidence for the incorporation of 14C-glycerol into lipids could not be obtained. The incorporation of 32P-orthophosphate into phosphatidic acid was increased by the addition of diglyceride, but not affected by that of glycerophosphate. These results suggest the possibility that phosphatidic acid is formed by the phosphorylation of diglyceride, but not by the acylation of glycerophosphate.
