Evaluation of Endocrine Disrupting Activities of Metabolites of Nonylphenol Polyethoxylates by Yeast Two-Hybrid Assay.

Metabolites of nonylphenol polyethoxylates (NPnEOs) such as nonylphenol (NP), nonylphenol monoethoxylate (NP1EO), nonylphenol diethoxylate (NP2EO) and nonylphenol triethoxylates (NP3EO), nonylphenoxyacetic acid (NP1EC), nonylphenoxyethoxyacetic acid (NP2EC), and brominated nonylphenol monoethoxylate (BrNP1EO) (collectively referred to“NPE-c”) were examined for their endocrine-disrupting activities by yeast two-hybrid assay. Among the tested NPE-cs, only NP showed a significant estrogenic activity and was 2.1×10-4times as potent as 17β-estradiol (E2) . Estrogenic activities of E2 and NP were 300 times (E2) and 17.5 times (NP) lowered by the incubation with S9mix. In contrast the metabolic activation of NP1EO with S9mix caused the occurrence of a significant estrogenic activity and 6.5×10-6times as potent as E2. It has been suggested that metabolic activation of NPE-c with S9mix have possibility to generate compounds which caused higher estrogenic activities. None of the progestrogenic, androgenic and thyroid hormone activities were found in the tested NPE-cs.
