Ureid (I) is hydrolyzed by NaOH into urea which reacts with dimethylglyoxime-thiosemicarbazide to give a violet-red color (λmax absorption at 530mμ) in an acidic medium.<BR>A new spectrophotometric method for (I) based on this color reaction was established and applied to the determination of (I) in pharmaceutical preparations.<BR>The influences of 47 compounds on the determination were in investigated, among which those by ureid, riboflavine, ascorbic acid, aspartic acid, acetaminophene etc. could be eliminated by chloroform extraction from an alkaline or acidic medium.<BR>The analytical procedure was as follows. Take the sample containing several mg of (I) in a separatory funnel, 10ml of water and 10ml of 2% NaOH solution, and extract 3 times with 15 portions of chloroform.<BR>Combine the chloroform extracts and evaporate on a water bath. Dissolve the residue in 10% NaOH solution and make up exactly to 50ml.Transfer a 2ml aliquot into a 30ml glass-stoppered test tube, and boil in a water bath. After cooling add 5ml of dimethylglyoxime-thiosemicarbazide mixed solution, and warm for 30 min. at 90°C. Cool to room temperature in running water, and make up with water to 25ml. The absorbance at 530 mμ (ET) is measured against a reagent blank. At the same time, the absorbance of the standard solution (ES) is determined and the amount of (I) is calculated by ET/ES.
