Description
The effect of the flow rate and CO2 concentration of air on the pH of medium in a jar fermentor culture system was studied. The CO2 concentration inside the jar fermentor was monitored and controlled by a micro-computer. Temperature and pH of the liquid medium were also monitored.<br>In the experiment, air mixed with CO2 gas flowed into the jar fermentor and it agitated the liquid medium. CO2 concentration were maintained at 400, 1000, 2000 and 3000ppm.<br>The following results were obtained: (1) The pH of the medium decreased as CO2 concentration or the flow rate was increased. The pH decreased more and faster in the orders of distilled water, MS1/2 medium, MS1/2 medium with sucrose 20g/l. (2) The pHs measured at aeration state, regardless of the CO2 concentration, were lower than those calculated at the equilibrium state without aeration, which clearly showed the effect of the flow rate on the pH of the media. (3) The measured pH of MS1/2 medium at aeration state showed to be nearly equal to that calculated at the equilibrium state minus the pH decrease due to the flow rate. (4) In case of culturing the Cymbidium PLB in this system, the pH of the medium decreased to 5.0 in 2 days after the start of the culture, and changed little in 22 days after that.
