Description
Chronic diabetic monkeys (Macaca fuscata) were produced with streptozotocin and they were kept without insulin treatment up to 30 months.
Hyperlipidemia in diabetic monkeys was characterized by hypercholesterolemia and hypertriglyceridemia. The major subfraction for cholesterol increase was attributed to LDL, but not HDL. No change of HDL2 cholesterol content was also observed. TG increased in every lipid subfractions, but mostly in VLDL fraction. Apoprotein B increased by 2.8 fold while apoprotein A1 increased by 30% in chronic diabetic monkeys.
By electron microscopy of abdominal aorta, intimal detachment, deposits of foam cells and proliferation of smooth muscle cells were observed in diabetic monkeys with duration more than 1 year. Significant thickenning of capillary basement membranes of muscle and renal glomeruli was also observed in these monkeys.
In conclusion, insulinopenia induces increased LDL and VLDL with increase of apo B. No significant change of cholesterol contents is noted in HDL subfractions.
Early lesions of both macro-and micro-vascular complications have been produced by metabolic derangement resulting from insulinopenia.
