Analysis of LDH isoenzyme patterns in cell lines of the small cell carcinoma (lung) and retinoblastoma.

Abstract

The LDH isoenzyme pattern of human lung cancer and retinoblastoma cell lines was studied by enzyme cytochemical and electrophoretical procedures. Seven small cell carcinoma, two adenocarcinoma and two retinoblastoma cell lines were used in this study. By the urea inhibition test of LDH enzyme cytochemistry, none of small cell carcinoma and retinoblastoma cell lines showed any decrease of LDH staining. However, the intensity of LDH stain decreased in both cell lines of adenocarcinoma by the urea inhibition test. By the electrophoretical analysis of LDH isoenzyme pattern both small cell carcinoma and retinoblastoma cell lines showed almost the same LDH isoenzyme pattern with the peak of LDH II or III, and their M/H ratio of LDH was 0.86±0.16 and 0.92±0.15, respectively. On the other hand, the adenocarcinoma cell line revealed high activity of LDH IV and V, and subsequently showed increased M/H ratio of LDH. In short, the LDH isoenzyme pattern of small cell carcinoma is very similar to that of retinoblastoma rather than that of adenocarcinoma. Therefore, these results may indicate a common biological nature between small cell carcinoma and retinoblastoma as a member of neuroendocrine tumors.
