In an attempt to explain the origin of positive endocochlear potential (EP) the endolymphatic surface was exposed to agents (barium, valinomycin and nystatin) which are known to alter cellular membrane conductances specifically in other tissues. This was accomplished by the use of a technique for perfusion of scala media. Application of barium to the endolymph caused the EP to increase in normal animals and exhibited no effect on the EP in Waltzing guinea pigs. On the contrary, both valinomycin and nystatin perfused in the scala media caused a decline in the EP to some extent. These results could support the cellular model in the marginal cell of the stria vascularis implied by Sellick and Johnstone (1975) ; the presence of driving force mediated by Na/K ATPase in the basolateral membrane and additional potassium transport mechanism in the luminal membrane (the endolymphatic surface).
