Endotoxin was extracted from Haemophilus influenzae type b by Westphal's phenol water method (lipopolysaccharide: LPS) and Boivin's method (Boivin's antigen). Forty guinea pigs were inoculated with 10ug/ml solution of LPS, and twenty with 80.ug/ml suspension of Boivin's antigen by transmeatal injection (on day 0). These were sacrificed from day 1 to day 23.<BR>Massive serous effusion filled tympanic bullae on day 1 and 3, and thereafter effusion decreased gradually, and it could hardly be seen on day 10. The pathological changes of mucosa were marked interstitial edema, dilated capillaries, high epithelium and intracellular edema, and they gradually subsided until day 23. The specimen irrigated with carbon in water suspension showed increase of permeability of capillaries. The specimen dyed with PAS-Alcian blue showed increase of mucous secretion.<BR>We conjectured that major pathogenic factors of the effusion were transudation and injury of epithelium which disturbed mucociliary transport activity, and increasing of secretion somewhat participated in inducing the effusion.
