The reaction of epidermis to trypsin before glutaraldehyde fixation differs between 4°C and 37°C (Br. J. Derm. 102: 75). On the other hand, the dermo-epidermal junction reacts differently at 37°C to trypsin before and after glutaraldehyde fixation (Rinsho Hifu 25: 677). It has been suggested that desmosomes in the granular layer are chemically different from those in the basal and spinous layers. In the present study the granular layer treated with trypsin at 4°C was observed before and after glutaraldehyde fixation. In both situations the cementing substances of desmosomes were digested similarly indicating that there are no chemical differences of cementing substance of desmosomes between the granular and basal-spinous layer as far as the glutaraldehyde fixation is concerned.
