The effects of sodium vanadate, an inhibitor of Na-K ATPase known as a potent diuretic agent, were studied on New Zealand rabbit kidneys, using clearance and isolated tubule microperfusion techniques.Addition of 20μM Na3VO4 (high dose) to the rabbit increased urine flow rate (V), UNaV and FENa, but had no effect on GFR, UKV and FEK. Addition of 10-6 M Na3VO4 (low dose) to the mucosal side inhibited reversibly the transepithelial electrical PDs in S1 proximal tubule, medullary thick ascending limb of Henle and distal convoluted tubule, but had no effect on PDs in S3 proximal tubule and cortical thick ascending limb of Henle. Further, 10-4 M Na3VO4 (high dose) added to seroaal side had no effect on PDs in these nephron segments. We conclude that vanadate suppresses the PDs in various nephron segments, more effectively from the mucosal side than the serosal. Inhibitory effect of vanadate in low concentration on PDs, hence probably the NaCl reabsorption, in these segments supports a possibility that vanadate may be one of the physiological regulators.
