A survey of mite allergens (Der 2) derived from mite bodies and numbers of mites was conducted to estimate mite infestations in the school environment in July, 2005. A vacuum cleaner was used to collect 146 dust samples from floors and bedding in 29 primary and secondary schools in Minato Ward, Tokyo. The collected house dust was extracted with phosphate buffered saline and the supernatant was used to measure Der 2 by an enzyme-linked immunosorbent assay (ELISA). The mites in the sediment were then isolated by the saturated saline flotation method. Only six samples (4%) from five schools exceeded the Standard Value of School Environmental Sanitation (100 mites per square meter). However, 56 of 146 samples (38%) exceeded 100 mites per gram dust, a WHO threshold for the development of IgE antibody and asthma, and 15 samples (10%) exceeded 500 mites per gram dust, another risk factor for acute attacks of asthma advocated by the WHO. Four suborders and 14 families including eight species were identified. Genus Derma-tophagoides including Dermatophagoides farinae Hughes and D. pteronyssinus (Trouessart), dominated at rates of over 90% in carpets, wooden floors and bedding. From these results, 100 Dermatophagoides mites per square meter were equivalent to 300ng of Der 2 in carpets, 210ng on wooden floors, 1,200ng in Tatami mats and 530ng in bedding. It was concluded that the Standard Value of School Environmental Sanitation was less strict than the WHO International Standard for mite allergen exposure.
