Newborn infants are protected against bacterial and most viral infections by the perinatal transfer of immunity from mother. Cellular immunity of neonates also provides some important protective roles in this stage. This work was undertaken to evaluate the inununologic status of SFD and prolonged pregnancy using Wistar rats.<br>SFD rats were obtained by the actinomycin D administration in the middle gestational stage (11th and 15th day) and also by the blocking of utero-placental blood flow using microspheres in the late gestational stage (18th day). Prolonged pregnancy was produced by consecutive injection of progesterone.<br>The results were obtained as follows:<br>(1) By followingu p the fetal uptake of 125I-IgGf rom the maternal circulation, it has been shown that the transfer of IgG occured entirely across the placenta and remarkably increased since 17th day of gestation and rapidly decreased since 23rd day.<br>(2) Fetal serum IgG level measured by the single radial immunodiffusion technique reached the maximum on the 22nd day of gestation and declined on the 23rd day.<br>(3) The most severe SFD was induced using the microspheres blocking technique in the late stage of pregnancy. However, injection of actinomycin D in the middle stage of pregnancy rather than in the late stage caused minimum IgG transfer. The author showed IgG receptors on the rat yolk sac endoderm by peroxidase-labeled antibody technique and demonstrated that their IgG binding ability was most affected by the placental damage in the middle gestational stage.<br>(4) In the neonatal period, labeled IgG transfer by suckling showed lower level in the Sill group and their serum IgG level didn't reach the level of normal group.<br>(5) In rats, PHA response was first demonstrable 20 days after birth (stimulation index>3) and reached the adult level at about 2 months of age.<br>(6) T cell function, assayed by PHA response (whole blood method), diminished in SFD induced by both of above methods, especially was severely supressed by actinomycin D administration on the 11th gestational day.<br>These results suggest that some SFD inducing factors may partially participate in some cases of perinatal immunodeficiency states.
