Cellular activity of the human amniotic epithelium was studied morphometrically using an autocytoanalyzer on eighty-five pregnant women. Significant differences in the morphometric findings were observed between the placental amnions and the reflected amnions after the 20th week of gestation.<br>After the 28th week of gestation, the total cell count in the unit area and the N-C areal ratio decreased remarkably in both amnions, although the decrement ratios were different in each amnions. On the other hand, cellular abnormalities such as binucleation and anucleation, intercellular vacant space, single cell area and number of sudanophilic granules in the cytoplasm increased remarkably in both amnions.<br>The age-correlated changes in normal amniotic epitheliums were examined by comparing the above mentioned findings with those of the intrauterine fetal fatalities. Consequently, these age-correlated changes in amniotic epitheliums were presumed to occur as the result of cell degeneration.<br>In the amniotic epitheliums of two cases with oligoamnios, severe degeneration was observed. However, in two cases of hydramnios with anencephalus, extended cell enlargement without degeneration was observed; in two cases of hydramnios with no fetal anomaly, hyperplasia of the epithelial cells was observed.<br>According to these findings, the cellular activity of the amniotic epithelium was presumed to contribute to the production and maintenance of the amniotic fluid. Therefore, the age-correlated degeneration of amniotic epithelium may cause a reduction in amniotic fluid volume.
