Abstract

The in vitro leukocyte adherence inhibition (LAI) assay against mixtures of autologous serum and suspected drug was used to detect the presence of cellular hypersensitivity in patients with suspected drug eruptions. None of the drugs used as antigens in this study, except isoniazid (INH), inhibited the adherence of normal leukocytes. Leukocytes from 18 of 21 patients with clinical manifestation of drug eruptions, to whom only a single drug was administered, produced LAI phenomena in response to the suspected drug Leukocytes from 3 of 8 patients, given 2 or 3 kinds of drugs, also produced LAI phenomena in response to the suspected drugs but not in response to other drugs Leukocytes from the control subjects, to whom the same drugs had been administered but who had not developed drug eruptions, as well as those from normal individuals, did not produce LAI phenomena in response to any of the drugs. This suggests that the LAI assay may be of use in the detection of causative drugs in patients with drug eruptions.
