Abstract

The pathophysiological condition of the mucosal epithelium with laryngopharyngeal reflux disease (LPRD) is still obscure. We attempted to make a model for acid exposure using a trachea excised from a guinea pig. We evaluated the electrical barrier function of this model using the Ussing chamber technique. First, we investigated the effect of apical acidity on the short-circuit current (SCC) of the tracheal model. From pH 2, SCC was significantly increased, which indicates impaired barrier function. The impaired barrier functions exposed by HCl were recovered after apical ZnCl2. In this model, lansoprazole had no effect on the impaired barrier function and pepsin had no effect on SCC. These findings help to provide a new strategy for LPRD therapy.
