Description

This new type of ampul was deviced for the preservation, transportation, and insemination of the bull spermatozoa.
The followings are the comparison of results between the ampul storage method and the usual test-tube storage method, with the special reference to the effects of anaerobous medium on the spermatozoa in the ampul, which results from the sealing of the opening of the ampul with gelatine whose melting point is bolow 38°C.
1. As to the survival period, the ampul method prolonged it to 49.71 hours in 50% of spermatozoa survival, and 36 hours in 30% of spernatozoa survival.
2. The decreasing of degree of motility of spermatozoa in storage was indicated by their survival and activity percentage. The chart shows that by ampul method the motility of the spermatozoa was maintained for about two days, as well as the time of collection.
3. In insemination test, the fertility percentage was 81% within 24 hours. 100% in 25-50 hours, and 75% in 51-75 hours and 50% in 90-125 hours.
The long distance transpcrtation test showed 85% fertility in both cases from Chiba-city to Hokkaido and Hiroshima Prefecture.
4. In Methylene blue reduction test, significant correlation existed between the number of spermatozoa, their activity and reduction time. Presumably it will contribute to the judging of the quality of the semen.
