Injection of 0.01mg of estradiol benzoate (EB) into egg yolk caused reduction in size of both right and left testes1). In this experiment, using the same experimental materials and design, dose of EB was doubled to examine whether estrogen sensitive period and response of left testes are dose dependent or not. Quail eggs for this experiment were obtained from autosexing lines and genetical sex of the embryos was readily distinguishable on day-16 of incubation. Single dose of 0.02mg of EB was given at different days of incubation and all embryos were killed at day-16 to inspect the gonads and duct system. According to the degree of asymmetry of the left and right testes, or to the feminization of the testes, the responses were classified into 4 grades. The injection of EB could increase the size of left testes to or over the controls, while the average size of right testes was decreased depending upon the time of hormone injection. Such changes of male gonads in relative size were similar to that seen in the female. In both sexes of embryos day-4 was the most susceptible period to EB. Mullerian duct was chosen as another parameter to estimate the feminization. By the changes of duct system, hormone treated embryos were classified into four groups. Although slight changes of gonadal shape occurred in all embryos, marked changes appeared exclusively in embryos which received injection in a day-0 to day-4 period. EB treatment after day-5 was practically without effect on the duct system.
