In an attempt to estimate the changes of protozoal polysaccharides contents after feeding different diets, two sheep were fed either high protein diet (PD) containing 30g of milk casein or high starch diet (SD) containing 70g of potato starch. The rumen liquor taken through a rumen fistula at intervals of 2 hours was analyzed for characteristics of the rumen liquor and polysaccharides contents of the ciliates genus Entodinium. The results are as follows: 1) Maximum concentrations of the ruminal VFAs were manifested at 4 hours and 2 hours after the SD and PD feeding respectively. 2) The pH of rumen liquor ranged from 6.8 to 6.4 according to VFAs production rates in the rumen. 3) The ammonia-N concentrations of rumen liquor were high in the PD feeding period. 4) The protozoal glutamic-pytuvic transaminase (GPT) activity was always high in the PD feeding period and exhibited a considerable increase 2 hours after feeding. 5) When the SD was fed the total count of protozoa genus Entodinium increased almost twice as much as that in the PD feeding period. 6) The increase in polysaccharides contents of the ciliates 2 hours after the SD and PD feeding was 20 and 2mg per ml of rumen liquor respectively. The value in terms of mg per 105 protozoa was 3. 5 and 1. 2 respectively. During from 2 hours to 10 hours after feeding 21.5 (SD) and 3.2 (PD) mg of protozoal polysaccharides per ml of rumen liquor or 3.5 (SD) and 1.3 (PD) mg per 105 protozoa were decreased.
