Description
Fixation of CO2 by washed goat spermatozoa at 37°C under an aerobic condition was investigated. The results obtained are as follws. A large amount of radioactivity which implies the fixation of CO2 was detected in the acid soluble fraction of reaction mixture when spermatozoa were incubated with 14CO2 or NaH14CO3. Almost similar amount of 14C was incorporated either when pyruvate, lactate or glucose was used as substrate, but was less without substrate. Almost all of the 14C fixed by spermatozoa (98%) accumulated in the acid soluble fraction, and a small amount of it was incorporated in the protein fraction. About 79% of 14C incorporated in the acid soluble fraction was recovered in anionic fraction, 20% in cationic fraction, and only about 1% in neutral fraction. Approximately 95% of the 14C labelled compounds in the anionic fraction were present as fumalate (1.5%), lactate (21.2%), α-ketoglutarate (12.6%), malate (20.9%) and citrate (39.0%). Approximately 90% of the 14C labelled compounds in the cationic fraction were as aspartate (9.8%), glutamate (50.1%), glutamine (15.8%) and alanine (14.2%). Malic enzyme activity was found in the extract of washed goat spermatozoa, but pyru-vate carboxylase was not.
