For better utilization of pigskin collagen as a industrial material, it is important to elucidate the macromolecular structure and to find out better methods of solubilizing insoluble fraction which covers greater part of total collagens. In this study, the solubilization of insoluble pigskin collagen and the composition of the solubilizates were investigated in comparison with those of bovine insoluble collagen. When insoluble collagens were heated in the buffer solution of pH 7.5 at 60°C, both the collagens of 6-months old pigskin and calfskin were solubilized in the amount of about 60% for one hour heating and about 90% for 22 hours heating, while both the collagens of 3-years old pigskin and cow hide were solubilized in the amount by 30-40% less than the former two. The similar tendency was also observed in the case of solubilization with 2M calcium chloride at 20°C or 1M urea at 45°C. Therefore the dependency on age is very large. However, when insoluble collagens were treated with pepsin in advance of solubilization the dependency on age almost disappeared, thus the solubilized amount of collagen increased to more than 80% for both of the pigskins and more than 60% for both cow hide and calfskin. Acryl amide gel disc electrophoresis and Sepharose Cl-4B chromatography of the solubilizates indicated the increased proportion of α component of collagen subunits and the change was more remarkable in pigskin collagens than in bovine ones.
