Abstract

Heterogeneity of equine transferrin (Tf) protein was analyzed about 15 Tf phenotypes composing of D, F, H, O and R alleles by means of two-dimensional electrophoresis (2-D method).And five samples per each Tf type were used.Each heterogeneity of the Tf O and R types having slow running mobility was detected as eight spots (T1-T4 and T′1…T′4), because main-bands (T) and sub-bands (T′) of both Tf types were separated and had four spots with different pI value, respectively.However, each heterogeneity of Tf D, F and H types with fast running mobility was observed as four spots (1-4), because their T and T′ components were mutually overlapped due to the narrow mobility between both components.Although heterogeneity patterns within each Tf type were bassically same ones, some changes were observed in the number and density of spots.T and T′ components of the Tf types were changed respectively from four to one spot by treating the serum with1% ammonium ferrous sulfate solution (1: 10).From these facts, it was assumed that the heterogeneity of equine Tf protein by the 2-D method is a functional one which changes by binding with iron ion and so on.
