The toxicity of the unsaponifiable fraction in TF (a toxic by-product of rice oil rendering) was concentrated on fraction 2 (F2) by alumina column chromatography, and F2 contained very much Cl. This Cl was based on chlorinated biphenyl (sold as Kaneclor 400 or KC400). Therefore a toxic substance in TF would probably be KC400.<br>The authors, with the objective to confirm the toxicity of KC400, studied the toxicity on chicks and the distribution of KC400 in them. Besides, analysis procedure of KC400 in TF was improved.<br>The results obtained are summarized as follows:<br>1. As a result of feeding KC400 to chicks at 10, 50 or 200ppm in a practical type of ration for four weeks, the mortality of chicks fed 50 and 200ppm was 75% and 100%, respectively. Those fed 10ppm KC400 survived but were found accumulation of fluid in the pericardial sac and changes of the internal organs.<br>2. The toxic symptoms elicited by KC400 were all the same syndrome that chicks fed TF showed. And the mortality nearly coincided with that by KC400 in TF.<br>3. The levels of KC400 accumulated in each tissues were adipose tissues, liver and kidney in order of height.<br>4. "Short column" method could be adapted for analysis of KC400 in fats. Summarized results of I-III<br>The results of studies on the toxic principle in a certain by-product of rice oil rendering are summarized as follows:<br>1. A toxic by-product (TF) derived from a particular lot of a particular renderer was toxic on chicks, and chicks fed TF showed the syndrome which was closely resembled to that of so-called chick edema disease in U. S. A.<br>2. Toxic principle appeared in the unsaponifiable fraction of TF and was concentrated on the fraction which contained a large quantity of Cl by alumina column chromatography.<br>3. In gas-liquid chromatographic analysis, the chromatogram of the toxic fraction completely corresponded with that of chlorinated biphenyl (sold as Kaneclor 400, KC400) used as a heat transfer medium in rice oil rendering process connected with TF. TF undoubtedly contained KC400 and the content was calculated to be 1, 350ppm.<br>4. As a result of the test using chicks in order to investigate the toxicity of KC400, chicks fed KC400 showed the same syndrome that TF elicited. And the mortality of chicks fed 50 and 200ppm for 4 weeks was 75% and 100%, respectively. While the toxicity of KC400 was coincided with that of TF qualitatively and quantitatively.<br>5. The levels of KC400 accumulated in each tissues were adipose tissues, liver and kidney in order of height. KC400 tended to be electively accumulated in adipose tissues.<br>As stated above, the toxic principle in a certain by-product of rice oil rendering was exactly identified as chlorinated biphenyl (KC400, containing 48% chlorine), a commercial heat transfer medium.
