For the purpose to study on the nature of antigenic determinant of β-lactoglobulin (β-Lg), the antigenicity of β-Lg reacted either with urea, p-chloromercuribenzoate (PCMB), 2-mercaptoethanol (ME), iodoacetic acid (IA) or hydrogen peroxide (HP) was examined. β-Lg, which had been allowed to stand for 72hrs in 8M urea solution (pH6.6) or in 8M urea solution containing L-cysteine, formed polymers, but no, polymer was formed when β-Lg had been stored for 72 hrs in 8M urea solution containing N-ethylmalelmide. The antigenicity of the β-Lg treated with urea was compared with that of native β-Lg. Although 1.08 sulfhydryl groups per mol(mol wt 18000 daltans) of β-Lg had reacted with PCMB, no decrease in the antigenicity was observed in the β-Lg modified with PCMB. On the other hand, 75% of the antigenicity of β-Lg lost in the β-Lg which had been reacted with IA in alkalinic regions after treatments with ME, and 12.5% of the antigenicity lost in the β-Lg which had been reacted with IA or HP in an acidic region. From the results obtained, it was suggested that the antigenic determinant was not related to a free sulfhydryl group in native β-Lg.
