抄録

The concentration of free amino acid in red blood cells of horse were analysed, and the relation among the free amino acid types of red blood cells and concentrations of sodium, potassium and GSH, and enzyme activities of GPT and GOT were investigated. 1) In paper electrophorograms, horse red blood cells possesed three zones of ninhydrin-positive substances, and were classified into two types. Namely the higher relative density type of the first zone (more than 80%) designated basic amino acid positive type (BA+type) and the lower relative density type (less than 60%) designated basic amino acid negative type (BA-type). BA+type red blood cells have higher relative density of neutral amino acid zone than that in BA-type red blood cells also, but GSSG zone in BA+type red blood cells was not different from that of BA- type red blood cells. 2) BA+type red blood cells contained higher intercellular concentration of certain free amino acids particularly alanine, serine, threonine, tyrosine, lysine, ornithine and histidine, than that in BAtype red blood cells, and the concentration of total amino acid of BA+ type and BA-type was 14.8±1.2mM packed cells and 6.8±0.50mM packed cells, respectively. 3) The concentration of Na, K and Na+K in the red blood cells of BA+type were 41.7±1.2m equiv/l, 89±4.6 m equiv/l and 130±8.4m equiv/l, respectively were always lower than that in BA-type red blood cells which concentrations of Na, K and Na+K were 51.8±1.8m equiv/l, 104.0±1.2m equiv/l and 155.8±2.0m equiv/l, respectively. 4) GSH concentration in the red blood cells of BA+type (81±4.4mg/100ml) was not significantly different from that of BA-type red blood cells (79.7±3.3mg/100ml). 5) GPT activity in the red blood cells of BA+type (185.7±3.4 Unit of Reitman-Frankel) was not significantly different from that of BA-type red blood cells (190.7±1.9 Unit of Retiman-Frankel), and GOT activity in the red blood cells was also insignificant difference between basic amino acid types. Namely GOT activity of BA+type was 164.3±7.3 Unit of Reitman-Frankel and that in BA-type was 172.6±3.0 Unit of Reitman-Frankel. But both enzyme activities in BA+type red blood cells were a little lower than that in BA-type red blood cells.
