The contributions of ubiquitin, which has a regulatory role in the degradation of essential or abnormal intracellular proteins, to glucocorticoid (GL)-induced apoptosis in lympho- cytes in the bursa of Fabricius in the chicken were studied. A single injection of five-week-old chickens with dexamethasone resulted in significant decreases (p<0.01) in the levels of bursal ubiquitin 24 and 48h later. In a second experiment, in which ubiquitin levels were monitored over the first 24h after injection, ubiquitin showed a significant increase (p<0.05) 2h after injection and thereafter decreased with time. The level of bursal ubiquitin at 24h was significantly lower (p<0.05) than the initial level. It is suggested that 1) the transient increase in the levels of ubiquitin might be involved in DNA fragmentation, which may be one of the key events in the early phase of GL-induced apoptosis in bursal lymphocytes, and 2) the subsequent decrease in bursal ubiquitin might be reflected in the decline of lymphocyte viability in the bursa of Fabricius.
