In a previous study, we found that a ginkgo biloba extract (GBE) diet increased liver weight, enhanced gene expression of hepatic cytochrome P-450 in the liver, and attenuated the hypotensive effect of nicardipine. In this study, we examined the effects of GBE on the cardiovascular function of aged male Wistar rats (18 months) fed ad libitum GBE for 4 weeks. In addition, the effect of GBE for 4 weeks on the antihypertensive action of orally administered nicardipine was examined. Four weeks of feeding on GBE did not influence the heart rate and systolic and diastolic blood pressure of the aged rats. The hypotensive effect of nicardipine was significantly reduced in the rats fed GBE for 4 weeks, although there was no significant difference in the weight of the liver between rats fed the control diet and those fed the GBE diet. In the atria isolated from rats fed the control or GBE diets, there was no significant difference in isoproterenol-induced positive inotropic and chronotropic effects. Also, GBE feeding did not change acetylcholine-induced negative inotropic and chronotropic effects. However, a noradrenaline-induced contractile response of the rat aorta was significantly enhanced by GBE feeding. From these results, GBE may reduce the therapeutic potency of nicardipine regardless of age, and this raises the possibility that GBE and drug interactions may occur clinically.
