Genotypes of κ-casein in 32 Japanese Shorthorn cows were identified in order to ascertain their effects on the calf-lactational performance of the dam. DNA was prepared from cow's blood. The PCR was carried out using 250ng of template DNA and Tth DNA polymerase with 35 cycles of amplification (denaturation at 93°C for 1 min, annealing at 55°C for 1min, extension at 72°C for 1 min). Synthetic oligonucleotides were used as primers: primer 1 (sense primer), 5'-CGCTGTGAGAAAGATGAAAGATTC-3'; primer 2 (antisense primer), 5'-AGATTCAAGGAGTA-TACCAATTGTTG-3'. Primer 1 was the 5' terminal sequence of exon 4, and primer 2 was 26 nucleotides in intron 4; therefore the DNA fragment containing the majority of exon 4 was amplified by PCR. After amplification, 780bp of PCR product was digested with Hind III and Pst I, and analysed by agarose gel electrophoresis. The genotypes of 32 cows were divided into 15 AA (47%), 15AB (47%) and 2 BB (6%). The milk peformance of 30 cows excluding BB cows, whose number was small, was statistically analysed by the least-squares mean. The result suggests that there is no significant difference in milk peformance between AA and AB cows.
