Diagnosis of Photosynthetic Functions of Grafted Tomato Seedlings with Chlorophyll Fluorescence Measurement Techniques

Abstract

The chlorophyll fluorescence measurement technique can be used for evaluating photosynthetic functions of plants without actually sampling the plants. In this study, we applied two types of chlorophyll fluorescence measurement techniques: pulse amplitude modulation (PAM) chlorophyll fluorescence measurement (MINI-PAM) and chlorophyll fluorescence imaging (CF Imager), to evaluate the health of grafted tomato seedlings. We monitored changes in Fv/Fm of seedlings incubated under different light conditions: Light-treated (10 h/14 h [light/dark]) and Dark-treated (continuous dark), for 10 days. Dark-treated seedlings showed slight wilting on day 10. Total chlorophyll content of Dark-treated leaves slightly decreased during the 10-day treatment, but Light-treated and Dark-treated leaves did not differ significantly. Fv/Fm measured with MINI-PAM was almost constant for 10 days in Lighttreated leaves, although Fv/Fm in Dark-treated leaves significantly decreased. Similar results were obtained with CF Imager. Absolute values of Fv/Fm measured with MINI-PAM were significantly higher than corresponding Fv/Fm values measured with CF Imager throughout the experiment. This result was probably due to CF Imager underestimating Fv/Fm because the imager’s saturation pulse light intensity was not strong enough to saturate the plant’s photosynthetic electron transport reaction. And the decrease in Fv/Fm of Dark-treated leaves was larger with CF Imager (60%) than with MINI-PAM (6%). Part of this difference could be attributed to changes in leaf angles caused by wilting. The results suggest that Fv/Fm imaging does not provide accurate Fv/Fm values but has greater sensitivity than MINI-PAM Fv/Fm measurement for detecting subtle changes in health of grafted tomato seedlings.
