Abstract

UDP-D-galacturonic acid (UDP-GalUA) is a donor of galacturonic acid in pectin. This compound is not commercially available, and its preparative method with high efficiency is needed. UDP-GalUA pyrophosphorylase involved UDP-GalUA synthesis has been neither purified nor cloned. The UDP-sugar pyrophosphorylase from Pisum sativum possesses a broad substrate specificity. Here, we investigated UDP-GalUA pyrophosphorylase activity of this enzyme and applied it to the preparation of UDP-GalUA.<br>The UDP-sugar pyrophosphorylase catalyzed the formation of UDP-GalUA and pyrophosphate from GalUA 1-phosphate and UTP, showing that this enzyme has UDP-GalUA pyrophosphorylase activity. The enzyme had optimum pH of 6 and the Km values for GalUA 1-phosphate, UTP, UDP-GalUA, and pyrophosphate were 2.3 mM, 1.2 mM, 0.70 mM, and 1.3 mM, respectively. In the presence of inorganic pyrophosphatase, the enzyme produced UDP-GalUA in 91 % yield on a preparative scale. Thus this UDP-sugar pyrophosphorylase is useful for the highly efficient production of UDP-GalUA.
