Metabolism of 3,4'-O-isopropylidene pyridoxine (3,4'-IPP) and 4',5'-O-isopropylidene pyridoxine (4',5'-IPP) was studied in rats. Following an oral or intraperitoneal administration of 3,4'-IPP, urinary excretion of pyridoxine (PN) was about 2~5% of the administered dose at mole base. About 20% of orally administered 4',5'-IPP were excreted as PN in the urine, but about 1~2% were detected as PN in the urine after an intraperitoneal injection. A metabolite of 4',5'-IPP in the urine was separated and purified by chromatographic techniques. The metabolite was assumed to be 4',5'-IPP 3-sulfate by measurement of Rf values on thin-layer chromatography and ultraviolet absorption spectra, identification of acid hydrolyzed product and detection of sulfate ion after acid hydrolysis.
