Abstract

This paper describes anisotropic spherical collagen hydrogel microparticles (〜100 μm in diameter) for single-particle co-culture system. We fabricated a quantity of uniform-sized collagen microparticles easily by ejecting micro-droplets containing collagen solution and sodium alginate into calcium chloride solution under high centrifugal gravity by using centrifuge-based microfluidic device composed of theta capillary, capillary holder, and microtube. We successfully demonstrated in encapsulating differently-labeled NIH/3T3 cells in each hemisphere at high viability by using our collagen Janus microparticles. We believe that our collagen Janus microparticles could be available to control differentiation of stem cells in a heterogeneous microenvironment at single-particles and to co-culture various cells in different ECMs with massive and quantitative analyses.
