Abstract

Effects of SH-compounds and their disulfides on rat hepatic hexokinase and glucokinase in vivo and in vitro were studied. 1) In vivo experiments : The time course of variation of enzyme activity by administration of 200 mg/kg of 2-mercaptopropionylglycine (MPG), 200 mg/kg oxidized MPG, 200 mg/kg of oxidized glutathione intraperitoneally, 3 g/kg of glucose orally, and 0.2 IU/head of insulin intramuscularly to normal and alloxan-diabetic rats was examined. From testing normal rats, glucokinase activity reached the maximum 1 hr after administration of MPG, and 3 hr after administration of glucose. For 3 hr after administration of insulin, the enzyme activity was the same as control, and then gradually increased with time. In disulfide compounds, oxidized MPG did not change the enzyme activity, but oxidized glutathione inhibited glucokinase activity by about 50% 1 hr after administration. Time courses of glucokinase after administration of effectors to alloxan-diabetic rats resembled those in normal rats. Hexokinase activity in both normal and alloxan-diabetic rats was hardly affected. 2) In vitro experiments (addition to the medium tested) : MPG, glutathione, and cysteine apparently, and oxidized MPG and cystine slightly increased both activities of hexokinase and glucokinase prepared from normal rat liver in concentrations from 1×10-3 to 1×10-7M. Oxidized glutathione strongly inhibited enzyme activities in concentrations from 1×10-3 to 1×10-5M, but activated in concentrations from 1×10-5 to 1×10-9M. Mechanisms of the action of SH compounds and their disulfides on hexokinase were discussed.
