Serum levels, urinary and biliary excretions, and tissue distribution of ticarcillin (TIPC) were, determined after a single intramuscular dose of 20 mg/kg in rats, rabbits and dogs. Marked differences in serum levels and half-life of TIPC were found among different species of the animals used. Serum levels of TIPC in dogs were clearly dose-related after single intramuscular doses of 20, 40, and 80 mg/kg. The urinary and biliary recoveries and tissue distribution were similar to those of carbenicillin.<BR>Serum levels and urinary excretion of TIPC after a single intravenous dose of 1 g were determined in 6 healthy volunteers, and the mean serum level peaked at 91. 8 μg/ml 15 minutes after injection. The 8-hour urinary recovery of TIPC and its penicilloate averaged 79. 7 and 14. 8% respectively. The serum levels of TIPC in the 6 volunteers were analyzed using a two compartment open model, and the pharmacokinetic parameters were calculated to simulate serum levels of the antibiotic after drip infusion of different doses or various periods.
