Synergistic activity of bacmecillinam (BMPC, KW-1100) and amoxicillin (AMPC), in the combination ratio of 5:1, 1:1 and 1:5, was demonstrated in vitro and in experimental mouse protection tests infected with E. coli F-2519, S. marcescens T-55 and E. aerogenes F-1949.<BR>E. coli F-2519 showed MIC of 25 and 200μg/ml against MPC and AMPC, respectively, with the MICs between 0.39 to 0.78 μg/ml when the two drugs were combined. ED50 of BMPC and AMPC was >4000 and 193mg/kg alone in the mouse protection test infected with the organism, but the ED50s were lowered to 49.9-168 mg/kg when the two drugs were administered in combination.<BR>S. marcescens T-55 was resistant to MPC and AMPC with MIC of>400 and 100 μg/ml, respectively. In combination with the two drugs, the MICs were distributed between 6.25-50μg/ml. ED50 of BMPC and AMPC were>2000 and 159 mg/kg alone, but the ED50s changed in a range of 75. 3-329mg/kg in combination, too.<BR>E. aerogenes F-1949 exhibited MIC of 3.13 and>400 μg/ml against MPC and AMPC. In the combination studies, the MICs were lowered to 1.56-3. 13μg/ml. Separately, neither of the two drugs were effective in mouse protection test, but the ED50s of 91.6-280mg/kg were obtained in combined administration.
