Description
Etoposide is a semi-synthetic derivative of podophyllotoxin which has antitumour acti-vity is sneveral experimental and human malignancies.<br>Pharmacokinetic study of etoposide is a useful tool to rationalize and improve the clinical efficacy of the drug. Therefore, a convenient method for determining the plasma concentration of etoposide is essential for pharmacokinetic study of the drug.<br>Authors developed a modified high performance liquid chromatographic (HPLC) method for determining etoposide in plasma. Plasma samples were pretreated with a solid-phase extraction procedure.<br>Pharmacokinetic behaviors of plasma etoposide were investigated after 30-60 min infusion in 9 patietns. The β-phase elimination half-life, total body clearance and β-phase volume of distribution were 5.8±3.4hr, 0.49±0.21 ml/min/kg, and 0.19±0.06 l/kg, respectively. Result of the recent clinical pharmacokinetic study shows that the modified HPLC method is very sensitive and selective for monitoring the therapeutic etoposide levels in clinical research.
