Elongation of Myc. lepraemurium in vitro was first discribed by Hart and Valen- tine. This method is certainly interesting in the studies of cultivation of Myc. leprae- murium in cell free media, but this has two disadvantages; centrifugation for collecting the bacilli and deformation of the bacilli caused by centrifugation. In order to improve these disadvantages, therefore, a slide culture method which has been routined for detecting the growth of tubercle bacilli was introduced into observation of elonga- tion of Myc. lepraemurium in vitro. The results obtained here demonstrated that the slide culture method was a simple and available method rather than the original method of Hart and Valentine and elongation of Myc. lepraemurium reported by Hart and Valentine was recon- firmed by this method, that is, elongation occurred especially at pH 6 of medium. In the experiments, a relationship between elongation and infectious activity was studied. According to the results obtained, the following relation might be presumed; elongation of the bacilli is distinctly a first step of fission of the bacilli until 15 days' incubation in the medium at pH 6, but thereafter elongation might indicate degene- rative deformation of the bacilli, because infectious activity of the bacilli was kept until 15 days' incubation in the medium at pH 6 and no infectious activity was remained after 30 days' incubation.
