In this study, action of luteinizing hormone releasing hormone analogue (LH-RH-A), human chorionic gonadotrophin (hCG) and pregnant mare serum gonadotrophin (PMSG), response to the hormones, and requisite events for starting ovarian cyclic activity by treatments with the hormones were examined in ovarian quiescent cattle. On the results obtained a hormone therapy, which was expected to be effective, was devised and efficacy was tested. Furthermore, pathological condition of the disease was investigated through analyzing reactions to the hormones and by measuring plasma luteinizing hormone (LH) level in affected ones. Followings were clarified. 1) LH-RH-A (100400μg) induced ovulation about 36 hr after treatment through causing LH release. 2) hCG(7506, 000 IU) induced ovulation about 36 hr after treatment without LH release. 3) PMSG (5004, 000IU) stimulated follicle(s) to develope, mature and secrete estradiol-17β(E2), and so that brought on ovulatory LH surge about 40 hr after treatment. Ovulation was induced consequently about 70 hr after treatment. 4) The corpus luteum (CL) that developed after induced ovulation (induced CL) was hypoplastic and short-lived (Type I) on LH-RH-A treatment. On hCG treatment, Type I and well-developed but short-lived CL (Type II) were formed. On PMSG treatment, Types I and II and normal CL (Type III) were found in the CL. 5) For initiating ovarian cyclic activity it was requisite that the induced CL was functional enough to elevate plasma progesterone level, and a follicle matured with active E2 secretion in accordance with regression of the CL. 6) PMSG (500 IU) was supplementaly dosed 6 days after LH-RH-A (200μg) treatment so as to stimulate maturation and active E2 secretion of a follicle developing along with regression of the CL for promoting ovarian cyclic activity. By the treatment ovaries started cyclic activity in most cases. 7) Although ovary, pituitary and hypothalamus were responsive to hormone stimuli, pulsatile LH release was not observed for more than 8 hr. It was considered that ovarian quiescence involved functional suppression of the hypothalamus, which presided over development and maturation of follicle by secreting LH-RH regularly, in pathology.
