The embryos which had been collected from 23 Japanese Black cows were equilibrated in 10% glycerol solution in one step method. They were then loaded into straws in the order of the sucrose solution, air, sucrose solution, air, glycerol solution with embryo, air and sucrose solution. The sucrose solution contained fluction V bovine serum albumin instead of bovine serum.<BR>Whenever each solution was loaded into the straw, extra fluid on the inner surface was absorbed by sterilized filter paper which avoided mixing of the different solutions used. Almost the whole length of the straw was placed in the freezing chamber and the tip was placed in the seeding chamber. Both chambers were separated by the heat insulater in order to avoid overcooling of embryo at the time of ice-seeding. Frozen straws were stocked in the liquid nitrogen for 2 to 7 months before embryo transfer.<BR>After the frozen straws were thawed, the viability of all the embryos were examined by a stereoscopic microscope and only morphologically good embryos were transferred. Some straws were used for the direct embryo transfer and others were used after the embryo was once removed from the original straw and then loaded into the new one (the indirect method). Heifers of 13 Japanese Short Horn cross bred, 12 Japanese Brown cross bred and 15 Japanese Black were employed as recipients. Embryo transfer was nonsurgically conducted 1 to 3 times for each recipient during late September 1985 to late January 1986.<BR>Pregnancy rate determined by rectal palpation was high in cross bred heifers compared with Japanese Black heifers (22/43 or 51.2% vs 6/19 or 31.6%). The direct transfer was superier to the indirect transfer in cross bred heifers (11/18 or 61.1% vs 11/25 or 44%) and in Japanese Black heifers (3/6 or 50% vs 3/13 or 23.1%). Skilled operaters obtained higher pregnancy rate than unskilled ones in cross bred heifers (21/37 or 56.8% vs 1/6 or 16.7%) and in Japanese Black heifers (5/15 or 33.3% vs 1/4 or 25%).
