F1/1 was isolated from a blastocyst obtained from a C57BL/6 female mated with a CBA male as a pluripotent murine embryonic stem (ES) cell line. The proliferation and differentiation of F1/1 cells were examined by culturing with different feeder cell layers or with leukemia inhibitory factor (LIF). The proportions of the colony formation and of the alkaline phosphatase positive colony of F1/1 cells were higher when they were cultured for 3 to 7 days with a primary embryonal fibroblast cell layer than the STO cell layer. These feeder cell layers are indispensable for inhibiting differentiation of F1/1 cells during culture. F1/1 cells were cultured through 15 passages with LIF without feeder cell layer; the cells were maintained undifferented and 68% of them had normal karyotype (38+ XY). However, no youngs were obtained when chimaeric embryos which were produced between 8-cell stage embryos and F1/1 cells maintained with LIF were trasferred to recipients, while youngs were obtained from 50 to 88% of chimaeric embryos with F1/1 cells maintained in the culture with feeder cells.
