The method to grow pine saplings infected with T. matsutake was devised in order to inoculate the fungus on adult trees and to cultivate it in the field or in greenhouse. The saplings of Pinus densifiara and some other pine species were planted along the margin of the colonies (Shiros) of T. matsutake in Pinus densifiora stand. The formation of mycorrhiza on the roots of saplings was recognized between 8 to 12 months after planting. The mycelium on the infected roots of saplings survived for at least 6 months after transplantation into the adult pine stand. The following findings are indicated by this study. 1) The young and actively growing Shiros of T. matsutake should be selected and traced precisely before the work begins. 2) The shrubs around Shiros used as the inoculum must be clearly cut off to make the saplings grow well. 3) The saplings grown in the nursery and which bear abundant fine roots are more suitable for the formation of the mycorrhiza than the ones grown on the forest floor. Moreover, the saplings grown in sterilized and infertile soil and with few mycorrhizae seem to be best for this purpose. 4) Before planting, the roots of saplings must be washed repeatedly after trimming off fine roots and other mycorrhizae. The sterilization by fungicides is unnecessary. 5) The proper term for the planting of saplings to the margin of Shiros is early spring from February to March, and that transplanting to other sites is best from November to April. 6) The typical mycorrhiza is not formed on the roots of pine species except for Pinus densifiora.
