The effects of prostaglandin E1 (PGE1) on mucin secretion and mucosal lesions in rat stomach induced by indomethacin and cold restraint were investigated.<br>The mucus content was measured by the alcian blue binding method. Intravenous infusion of PGE1 (10μg/kg) for 1 hour increased the total amount of mucosal and adhesive mucus at the rate of 14.2%. The rate of increase in the amount of adhesive mucus on the surface epithelium was 16.7%. Both treatment by oral administration of indomethacin (60mg/kg) and by cold restraint at 4 °C for 1 hour produced acute mucosal lesions and decreased the mucus contents in gastric mucosa at the rate of 17.8% and 16.1%, respectively. However, the mucosal lesions and decreases in mucus content were markedly inhibited by the administration of PGE1 (10μg/kg) for 1 hour infused simultaneously with the above treatments.<br>These results suggest that a concentration of PGE1 so low that it hardly inhibited gastric acid secretion could induce an increase in mucin secretion and prevent ulcer formation in rat stomach.
