Some Properties of Inhibitors of Plant Virus Infection Occurring in the Leaves of French Bean

Abstract

Fractions, S1 and S2, separated from crude extracts of French bean leaves (var. Kairyo-Otebo), by precipitation with 0.5 saturated ammonium sulphate and Sh1 and Sh6 subsequent heating of S2 at 100C for 1 or 6 minutes were tested for their inhibitory effects on the infection of TMV to French bean, Nicotiana glutinosa, Chenopodium album and tobacco. Each fraction showed about 80% inhibition when assayed on French bean leaves but less inhibitory effect on N. glutinosa. The inhibitory substance was heat stable at 100C for 6 minutes. When S2 fraction was dialysed against phosphate buffer for 24 hours, the activity was markedly decreased. The mixture of TMV and S2 was easily separated into two fractions P1 and P2 by Sepharose 2B gel filtration; P1 contained the high molecular weight substances including TMV, while P2 low molecular weight substances which were contained in S2 fraction. When TMV was again mixed with P2 fraction and inoculated to N. glutinosa, the number of lesion decreased to 123 as against 400 with TMV only. It is indicated that the inhibitory substance could be a relatively low molecular weight, which is diffusiable in Sepharose 2B gel. Results of gel filtration chromatography, using G200, G100 and G50 suggested that the partially purified inhibitor (Sh6) contained several molecular sizes of compounds. Fraction P1 by G50 seemed to contain a mixture of high molecular weight compounds between 104-2×105MW, and P2 a low molecular substance which was eluted together with sodium fumarate added as a marker. Inhibitory effect of Sh6 on TMV infection was tested using French bean, N. glutinosa, C. album and tobacco. The number of lesion on tobacco leaves was counted after visualizing the lesions by heat treatment. The inhibitory effect was highest on French bean laeves.
