New benchmarks were developed for differentiating Streptomyces scabies, S. acidiscabies and S. turgidiscabies, that cause potato common scab. Secondary metabolites of the pathogens were extracted directly with methanol-diethylether (2:1, v/v) or acetonitrile from a solid Bennett's medium after 5 days of culture at 25°C. The extracts were spotted onto a precoated silica gel TLC plate (Merck Co., Si60, 0.25mm), which was then developed at 25°C for 90min, with chloroform-methanol-acetone-acetic acid-distilled water (55:20:4:4:2, v/v). The characteristic spots were detected by UV irradiation at 365nm, and the resultant chromatograms were recorded with a digital camera and analyzed with a personal computer. Because chromatograms are specific, three causal agents of potato common scab disease can be rapidly differentiated.
