An amphiphilic polymer, poly (allylamine) (MW=10000) containing 30 mol% hydrophobic poly (L-alanine) graft chains (MW=1065) (PAAgPAla) was prepared. PAAgPAla formed micelles in aqueous solutions. The size of the PAAgPAla micelle increased with increasing pH, reflecting ionization changes in the allylamine moiety in the main chain. In alkaline conditions, the PAAgPAla poly (L-alanine) graft chains were found to self-assemble owing to the absence of the electrostatic repulsions among the neighboring allylamine moieties, to yield β-sheet structure domains in the micelle. We investigated the molecular recognition properties of the PAAgPAla micelle at pH 10.0 using surface plasmon resonance measurements. Active domains composed from the poly (L-alanine) self-assembly in the micelle interact specifically with aqueous L-phenylalanine rather than with its D-isomer. The binding of L-phenylalanine to the PAAgPAla micelle exhibited Langmuir-type saturation behavior, with its affinity constant (K=3.4X105 M-1) being 7.2 times larger than that observed for its affinity for D-phenylalanine (K=4.7X104 M-1) The poly (L-alanine) domain in the PAAgPAla micelle was confirmed as a binding site for L-phenylalanine molecules.
