The purpose of this study is to establish the mechanism of generation of high tensile strength (T) in a tablet consisting of microcrystalline cellulose (MCC) particles with large L/D (L, length; D, width). An attempt was made to clarify particle orientation in a tablet by examining inner structure of the tablet using scanning electron microscopy and X-ray diffraction measurement. By observing the surface of MCC particles treated with cellulase, cellulose microfibrils were found to be oriented along longitude of the particle, and it seemed that the degree of crystallite orientation δ was correspondent with the degree of MCC particle orientation. Comparing δ among tablets consisting of MCC particles with different L/D, we found that δ increased with an increase in L/D and that the larger the L/D, the more highly MCC particles were oriented along the direction at right angles to the direction of compression, because MCC particles with a larger L/D might undergo a larger moment for rotating themselves so as to orient the MCC particles along the direction. We came to the conclusions that MCC particles with larger L/D could be easily oriented during compression, resulting in generation of a larger contacting inner surface among the MCC particles, so that the tablet consisting of the MCC particles with larger L/D exhibited larger T.
