Description
A trace amount of 2-mercaptobenzothiazole (MBT) in fish (carp) or in the aquarium water was successfully analyzed by high-speed liquid chromatography preceded by extracting MBT with methyl isobutyl ketone.In the case of fish, MBT was extracted by homogenizing the whole fish body with methyl isobutyl ketone in the presence of an antioxidant 2-mercaptoethanol.Successive cycles of freezing-thawing were sometimes found to be necessary in order to clarify the emulsion extracts.MBT in the aquarium water was extracted with methyl isobutyl ketone in the presence of 2-mercaptoethanol.An aliquot of the clear extract was concentrated in vacuo, dissolved in acetone, and was applied to a high-speed liquid chromatography equipped with a ODS column.MBT was eluted with a phosphate buffer (pH 3)-methanol (70/30) and was monitored at 320 mm.As low as 1 ppm and 0.01 ppm of MBT in fish and in the aquarium water, respectively, were quantitatively determined with a coefficient of variation of about 5%. By the present method, dibenzothiazyldisulfide, if any, should have been completely reduced to MBT by 2-mercaptoethanol and therefore was analyzed as MBT itself.
