The HPLC determination of cysteine, homocysteine and their disulfides in gelatin with isocratic elution and UV detection were investigated. Dithiothreithol (DTT) was used for reduction of disulfides and 4-(aminosulfonyl)-7-fluoro-2, 1, 3-benzoxadiazole (ABD-F), a fluorogenic reagent, was used for precolumn derivatization of thiols. ABD derivatized thiols were eluted isocratically and detected with not only fluorescence but also absorption at 375 nm. The detection limit of the analytes was about 2 ppm in gelatin using UV detection, whereas it was less than 1 ppm using fluorecence detection. The standard deviation was less than 2 ppm at 10 ppm level analysis for both detection methods. One column wash per day was suitable for the proposed conditions.
