Abstract

Bio–inspired nanomaterials (e.g., viruses, exosomes) that can deliver drugs efficiently to specific cells/tissues are more promising platform of drug delivery system (DDS) than conventional synthetic nanomaterials. Hepatitis B virus (HBV) is a hepatotropic DNA virus that infects human hepatic cells specifically. Inspired by the early infection machinery of HBV, we developed bio–nanocapsule (BNC), an yeast–derived nanoparticle harboring HBV L proteins on their surface. We currently identified two functional domains in the pre–S1 region of HBV L protein, such as membrane fusion domain (9–24 aa) and heparin–binding domain (30–42 aa). These domains might synergistically contribute to the efficient delivery of drugs to cytoplasm. Furthermore, we have been working on the application of exosomes for DDS. We proved that bovine milk–derived exosome is one of the promising platforms due to the capability for large scale production and safety in vivo. These exosome–based DDS may be useful for the efficient delivery of therapeutic RNA including siRNA and mRNA.
